Design Notes

The 30 base pair long gRNA spacer is complementary to a synthetic ProgRAM recording tape. It features an A to C mismatch in position 22 that is a target for deamination by ADAR2 deaminase domain. The sequence also includes a gRNA loop, that is essential for the dPspCas13b protein to recognize and bind the gRNA effectively.
